Wash protocol for Acetylcholine (AC-GEL) Column

AC-GEL Wash Protocol: 


Remove the enzyme column, and re-pack the precolumn. Collect wash solution directly from the column-end. NEVER pass effluent through the detector cell.
Wash as follows:
  • With distilled water for 60 min at the standard flow rate and in the forward direction.
  • With 100% methanol for 60 min at the standard flow rate reverse direction.
  • With distilled water for 60 min at the standard flow rate and in the forward direction.
  • Run mobile phase for 60 min at the standard flow rate in the forward direction
  • Connect to enzyme column and detector cell


Although it is possible to analyze more than a few thousand microdialysis samples between cleanings, it is only possible to analyze a few hundred samples of tissue homogenize. Thus, cleaning cycles will depend on what kind of samples are used.

.  Please ensure that the enzymatic column is detached as the organic solvent used to clean the column can permanently damage the enzyme activity.
